A comparative study on fungal laccases immobilized on chitosan BABT

The phenoloxidase enzyme laccase from the cultures of the Pleurotus ostreatus and Botryosphaeria sp. and a commercial laccase from Aspergillus sp. were immobilized on chitosan of pharmaceutical degree by adsorption followed by crosslinking. Different immobilization conditions in relation to the granulometry of support and amount of enzymatic laccase extract used were tested, aiming at reaching high enzymatic activity with the immobilized enzyme. Two different substrates, ABTS and DMP, were used for the determination of enzymatic activity. The highest enzymatic activity was obtained when 1.0mg/mL of the enzymatic laccase extract from Botryosphaeria sp. was used with 1.0g of support (200 mesh). Antifungal activity of essential oil isolated from Ocimum gratissimum L. (eugenol chemotype) against phytopathogenic fungi BABT

An investigation of antifungal activity of the essential oil obtained by steam-distillation (1.1% w/w) of the aerial parts of Ocimum gratissimum and of an ethanolic extract from the steam-distillation residue was carried out using the agar diffusion method. The results revealed that the essential oil inhibited the growth of all fungi tested, including the phytopathogens, Botryosphaeria rhodina, Rhizoctonia sp. and two strains of Alternaria sp., while the extract from the residue was inactive. The essential oil was subjected to TLC bioautography used to detect fungitoxic constituents. The compound that showed antifungal activity was isolated and identified as eugenol. Antimicrobial activity of essential oils from Pimenta pseudocaryophyllus and Tynanthus micranthus BABT

The present study describes the chemical composition and antimicrobial activity of essential oils obtained by hydrodistillation from the leaves of Pimenta pseudocaryophyllus (1.0% w/w) and Tynanthus micranthus (1.1% w/w). GC and GC/MS analysis demonstrated that eugenol was the only component in the T. micranthus essential oil (99.9%) and the major component in the P. pseudocaryophyllus essential oil (92.59%), which also presented methyleugenol, terpinen-4-ol, o-cymene and (E)-caryophyllene, among others. Xylanase production by Trichoderma harzianum rifai by solid state fermentation on sugarcane bagasse BJM

Sugarcane bagasse was used as substrate for xylanase production by means of a strain of Trichoderma harzianum Rifai isolated from decaying Aspidosperma sp. (peroba) wood. The bagasse was washed, dried, milled and wetted with minimal salts medium and the cultures grown at 28 ± 2ºC for 7 days. Two extraction methods were tested for enzyme recovery: (A) Tween 80, 0.1% (v/v), in physiological saline, and (B) 50mM sodium acetate buffer, pH 5.0, under agitation (180rpm) for 15, 30 and 60min. After a single extraction, both extraction methods recovered an average of 15U/ml of xylanase activity, independent on the time of shaking.
